Dapsone 100mg is classified under sulphonamide class of antibiotics and it is also called as diaminodiphenyl sulfone. It is frequently used to treat leprosy along with that it is also used to treat various skin diseases like Duhring’s disease, pyoderma gangrenosum, vasculitis and sweet syndrome.

Mechanism of action of Dapsone 100mg is not completely understood. It mainly acts by inhibiting the dihydrofolic acid synthesis by competiting with para-amino-benzoate for the active site of dihydropteroate synthetase. Dapsone shows anti-inflammatory effects, but as it is not steroid, it does not come under NSAIDs class.


Dapsone 100mg is indicated for the treatment of leprosy which is a bacterial infection that damages our skin and peripheral nerves in the hand and feet, mucosal layer of eyes, nose and throat. It should be taken empty stomach, but if stomach upset occurs, than it should be taken with food so as to reduce stomach irritation.


Dapsone 100mg is contraindicated in patients with

  • Severe liver diseases.
  • Anemia from pyruvate and glucose 6-phosphate dehydrogenase deficiency (G6PD).
  • Severe anemia.
  • Deficiency of blood enzyme cytochrome b5 reductase.

Drug interaction

  • Dapsone 100mg if co-administered with prilocaine or sodium nitrite increases the risk of methemoglobinemia.
  • Avoid using Dapsone 100mg and clozapine together as it may cause neutropenia and agranulocytosis that can increase the risk and/or severity of hematologic toxicity.
  • Dapsone 100mg should not be administered with didanosine as the latter causes the irregular absorption of Dapsone 100mg that leads to lowering of effects of drug.

Storage condition of Dapsone 100mg

  • Keep your medication tightly closed in the same container it came in.
  • Keep the drug out of reach of children.
  • Store the drug at room temperature and away from the excess moisture and heat.

Dose of Dapsone 100mg

Dapsone 100mg is available as a tablet dosage form.

Regular adult dose for:

  • Leprosy lapromatous- 50-100 mg once a day for 2-5 years.
  • Leprosy tuberculoid- 100 mg once a day for 6 months.
  • Dermatitis herpetiformis- 50-300 mg on a lifelong basis.
  • Pneumocystis pneumonia prophylaxis- 100 mg twice in a week on a lifelong basis.
  • Toxoplasmosis prophylaxis- 100 mg twice in a week on a lifelong basis.

Regular pediatric dose for:

  • Pneumocystis pneumonia prophylaxis- 2mg/kg per day up to 100 mg once a day.
  • Toxoplasmosis prophylaxis- 2 mg/kg/day up to 25 mg once a day.

Missed dose

In case if one dose is missed take that dose as soon as you remember. Skip the dose if it is time for next dose. Never go for double dosing in case of missed dosing.

Over dose

Over dosage of Dapsone 100mg is characterized by nausea, vomiting, seizures, easy excitability, and bluish discoloration of skin.

Side Effects of Dapsone 100mg

Some common side effects of Dapsone 100mg appeared in patients are mild skin rashes, weakness, fever, yellowing of skin, anorexia, backache, leg and stomach pain.

Rare side effects of Dapsone 100mg include redness, or dryness of skin, unusual bruises or bleeding, hair loss, mood swings, burning sensation of hand and feet and sore throat.

Precautions while taking Dapsone 100mg

  • Do not use Dapsone 100mg, if you are allergic to this drug.
  • Use of Dapsone 100mg may cause blurred vision, so avoid driving and other possible unsafe tasks.
  • Discuss your all other medication with your doctor as they may interact with the Dapsone 100mg.
  • Consult your doctor prior to using Dapsone 100mg, if you have a history of anemia or other blood disorders, liver disease, diabetic ketosis, serious heart or lung disease.
  • Avoid unnecessary or prolonged exposure of sunlight as Dapsone 100mg makes your skin sunlight sensitive.
  • Dapsone 100mg should be avoided in case of pregnancy as it passes into breast milk and can show undesirable effects on nursing infants.
  • Do not stop taking medication if you feel better as it is necessary to complete the full course of treatment for curing the infection completely.
